The plasma membrane Ca2+ channel in Leishmania species is a key component in the parasite's calcium homeostasis. It is functionally similar to L-type voltage-gated Ca2+ channels (VGCCs) but exhibits distinct pharmacological properties, including activation by sphingosine and miltefosine. It is considered a main target for miltefosine, and possibly other antileishmanial agents. Its unique activation profile makes it a promising target for selective therapeutic intervention against Leishmania parasites.
Activation of the channel by miltefosine leads to calcium influx, disruption of intracellular calcium homeostasis, and alkalinization of acidocalcisomes, contributing to parasite death.
