Store-operated Calcium Channels, notably those formed by Orai proteins, open when endoplasmic reticulum stores are depleted; they play critical roles in immune cell function and other signaling pathways[2]. The structure typically includes multiple transmembrane domains forming a central pore with selectivity filters that ensure preferential passage of divalent cations like Ca²⁺ over monovalent ions. Auxiliary subunits modulate trafficking to the plasma membrane and fine-tune biophysical properties such as gating kinetics and pharmacological sensitivity[4][3].
