Plasma membrane lipid raft

Molecular classification
Other (membrane microdomain)
01

Overview

Plasma membrane lipid rafts are **cholesterol- and sphingolipid-rich, highly dynamic microdomains** within the plasma membrane that compartmentalize cellular processes by serving as organizing centers for signaling assemblies, membrane trafficking, and protein sorting[1][3][5]. Their existence and precise definition are still debated, but substantial evidence supports their role in facilitating signal transduction, vesicle formation, axon guidance, immune responses, and pathogen entry[1][2][4][6]. Lipid rafts are implicated in health and disease across a range of physiological and pathological processes, but they *do not represent a discrete protein target* and thus are unclassifiable in the same sense as a single receptor or enzyme.

Mechanism of action

Cholesterol depletion/disruptors: Disruption of lipid rafts affects signaling by mislocalizing raft-associated proteins

03

Biological functions

Signal transductionMembrane trafficking and sortingAxon guidanceEndocytosis and vesicle formationImmune response (organization of key immune signaling events)Apoptosis and cell death signaling
04

Disease associations

Cancer (altered signaling and membrane protein distribution)Infection (entry platform for pathogens, altered immune signaling)Neurodegenerative disease (role in synaptic plasticity, axon guidance)Inflammation (modulation of reactive oxygen species and immune responses)Cardiovascular disease (involvement in endothelial function)Other (dysregulated raft-associated processes in various conditions)

Safety considerations

Non-specificity of targeting: Disruption affects many cellular processes, risking broad toxicityEssential functions: Lipid rafts are fundamental for cell signaling, trafficking, and survival in many cell types, raising safety concerns if targeted
