Plasmacytoid dendritic cells (pDCs) are a specialized subset of dendritic cells with a unique ability to produce large amounts of type I interferons (IFN-I), especially in response to viral infections. They act as key sentinels in the innate immune system, bridging innate and adaptive immunity through cytokine production and antigen presentation. pDC dysregulation is implicated in autoimmune diseases and cancer. Therapeutic modulation strategies include targeting TLR/MyD88 axis or IDO to reduce unwanted tolerance/immunosuppression. The transcription factor Spi-B is critical for pDC development/function.
