Plasmalogens are a unique class of ether-linked glycerophospholipids characterized by a vinyl-ether bond at the sn-1 position of the glycerol backbone [1, 14, 20]. They are essential structural components of biological membranes, especially in the brain, heart, and immune cells, where they maintain membrane fluidity and facilitate critical processes like vesicular fusion and neurotransmitter release [1, 9, 10, 21]. Beyond their structural role, plasmalogens serve as potent endogenous antioxidants, protecting cells from oxidative damage by scavenging reactive oxygen species (ROS) [1, 15, 21]. Deficiencies in plasmalogen levels are the primary cause of rare peroxisomal disorders such as Rhizomelic Chondrodysplasia Punctata (RCDP) and are strongly associated with neurodegenerative diseases like Alzheimer's and Parkinson's [3, 5, 8, 12]. Therapeutic strategies, known as Plasmalogen Replacement Therapy (PRT), utilize synthetic precursors such as PPI-1011 to bypass peroxisomal biosynthetic defects and restore physiological lipid levels in the brain and peripheral tissues [12, 13, 16]. Current clinical development focuses on these precursors to improve cognitive function, reduce neuroinflammation, and mitigate the progression of age-related neurological decline [6, 10, 21].
