Overview

Plasminogen is a glycoprotein zymogen that serves as the precursor to plasmin, a serine protease responsible for fibrinolysis. It is produced in the liver and circulates in the blood. Activation of plasminogen by tPA or uPA leads to the degradation of fibrin clots and other extracellular matrix proteins. Dysregulation of plasminogen activity can result in thrombotic or bleeding disorders, and it plays a role in cancer metastasis, inflammation, and tissue remodeling. Therapeutic strategies target this pathway through thrombolytic agents or antifibrinolytic therapies.

Mechanism of action

tPA and uPA analogs promote the conversion of plasminogen to plasmin, leading to clot dissolution. Antifibrinolytic agents inhibit plasminogen activation or plasmin activity, preventing clot breakdown.
