Overview

Plasmodium falciparum apical membrane antigen 1 (AMA1) is an integral membrane protein and a key molecule involved in the invasion of erythrocytes by malaria parasites. It is considered one of the leading vaccine candidates for malaria due to its essential role in parasite survival and its ability to elicit strong immune responses. AMA1 interacts with rhoptry neck protein RON2 on host cells, forming part of the moving junction complex necessary for successful erythrocyte invasion by merozoites. Disruption or inhibition of AMA1 function blocks parasite entry into red blood cells. However, high sequence polymorphism among field isolates poses challenges for broad vaccine efficacy due to strain-specific immunity.
