Plasmodium falciparum ATPase 4 (PfATP4) is an essential P-type cation translocating ATPase found in the plasma membrane of the malaria parasite Plasmodium falciparum. It functions as a Na+ efflux pump, maintaining low cytosolic Na+ concentrations critical for parasite viability and nutrient uptake. PfATP4 is a validated antimalarial drug target, with multiple classes of compounds inhibiting its function. Resistance to these drugs can arise from mutations in pfatp4, often with associated fitness costs.
Inhibition of PfATP4-mediated sodium efflux, leading to disruption of ion homeostasis and parasite death.
