The Plasmodium falciparum cyclic amine resistance locus (PfCARL), also known as Pf3D7_0321900, encodes a conserved protein implicated in the parasite’s resistance to several classes of antimalarial compounds, most notably the imidazolopiperazines such as KAF156 and GNF179. The exact physiological function of PfCARL remains incompletely understood, but it is predicted to play a role in protein folding within the endoplasmic reticulum and is localized primarily to the cis-Golgi apparatus in both asexual and sexual blood stages of P. falciparum. Mutations in pfcarl confer resistance to multiple structurally unrelated antimalarial compounds, representing an important mechanism for multidrug resistance.
