Pfs230 is a large, 363-kDa surface protein expressed on the gametes of Plasmodium falciparum, the parasite responsible for the most severe form of human malaria. It belongs to the 6-Cys family of proteins and is essential for the parasite's sexual reproduction within the mosquito midgut, where it mediates the adhesion of gametes to red blood cells and facilitates the fertilization process (PubMed: 31110283). Domain 1 (D1) of Pfs230 is the most immunogenic region of the protein and is the primary target for transmission-blocking vaccines (TBVs) currently in clinical development (PubMed: 32156742). When a mosquito ingests a blood meal containing Pfs230-specific antibodies—induced by vaccination or passive transfer—these antibodies bind to Pfs230 on the gamete surface, preventing the formation of zygotes or inducing complement-mediated lysis of the parasite (UniProt: P68874). This mechanism effectively halts the transmission of malaria from the human host to the mosquito vector, making Pfs230D1 a critical component of global malaria elimination strategies.
