Overview

Plasmodium falciparum gametocyte surface protein Pfs230 is a large (230-360 kDa) cysteine-rich protein belonging to the 6-Cys family, expressed on the surface of gametocytes and gametes (AJTMH, 2022; BioRxiv, 2025). It plays a critical role in the fertilization process within the mosquito midgut, specifically mediating male gamete fertility and the interaction between gametes and host erythrocytes (Singh et al., 2020; NIH, 2019). Domain 1 (Pfs230D1) is the N-terminal cysteine-rich domain and is the primary target for transmission-blocking vaccines (TBVs) due to its high immunogenicity and relative conservation compared to other domains (JCI, 2021; ResearchGate, 2023). Antibodies targeting Pfs230D1 interrupt the malaria transmission cycle by preventing the development of the parasite in the mosquito vector. These antibodies typically act through a complement-dependent mechanism to lyse gametes before fertilization can occur, making Pfs230D1 a leading candidate for malaria eradication efforts (Singh et al., 2020; Mesamalaria, 2024).
