Overview

The *Plasmodium falciparum liver stage* is the first stage of infection in humans, during which sporozoites infect hepatocytes, undergo extensive replication, and mature into thousands of merozoites that eventually infect red blood cells, leading to clinical malaria. This stage is asymptomatic but is a bottleneck and thus a promising target for vaccines and chemoprophylaxis. Key antigens and molecular complexes involved include CSP, LISP1, LISP2, EXP2, and the PTEX translocon, all of which are considered molecular targets within the broader "liver stage" context

Other names
Liver stage of Plasmodium falciparumexoerythrocytic stage (for malaria)pre-erythrocytic stage
02

Mechanism of action

Inhibition of *Plasmodium* development or survival in hepatocytes (e.g., disrupting mitochondrial function, interfering with metabolic pathways, causing parasite death before it reaches blood stage)

03

Biological functions

Infection establishmentParasite replicationEvasion of host immunityPreparation for erythrocytic (blood) stage infection
04

Disease associations

Infection (specifically malaria)
05

Safety considerations

Hepatotoxicity of certain drugs (e.g., primaquine and tafenoquine in G6PD-deficient patients)Lack of robust models for in vitro study, complicating drug discovery
06

Interacting drugs

Primaquine

07

Biomarkers

Liver-stage antigens, e.g. LISP1, CSP (circumsporozoite protein), EXP2, PTEX150 (used for monitoring infection or immune response in research, not clinical selection)
