Overview

Plasmodium falciparum meiosis-inducing protein 2-like protein (PfMei2) is a critical RNA-binding protein involved in the life cycle of the malaria parasite (UniProt: Q8I4V7). It is characterized by the presence of three RNA recognition motifs (RRMs), which are essential for its function in regulating RNA metabolism and gene expression during the liver stage of infection (PlasmoDB: PF3D7_1028100). PfMei2 is specifically required for the development of liver-stage schizonts; its absence leads to a developmental arrest that prevents the parasite from emerging into the bloodstream (Shindoh et al., 2007, PNAS). Because this protein is essential for the establishment of a systemic infection but is not present in the human host in the same form, it represents a high-value target for prophylactic antimalarial interventions. Research into PfMei2 focuses on its potential for developing live-attenuated vaccines and small-molecule inhibitors that can block the parasite before clinical symptoms arise. The protein's unique role in the transition from liver to blood stage makes it a focal point for understanding parasite maturation (Journal of Biological Chemistry, 2007).

Mechanism of action

Inhibition of PfMei2 prevents the maturation of liver-stage schizonts, thereby blocking the transition from the liver to the blood stage and preventing clinical malaria (Shindoh et al., 2007, PNAS).
