A Plasmodium falciparum sporozoite-infected hepatocyte refers to a human liver cell (hepatocyte) that has been invaded by the sporozoite stage of the malaria parasite P. falciparum. This is a critical early step in the malaria life cycle, occurring after an infected Anopheles mosquito injects sporozoites into a human host. The sporozoites travel via the bloodstream to the liver, where they invade hepatocytes and undergo development before releasing merozoites into the bloodstream, initiating symptomatic blood-stage infection. The invasion of hepatocytes by P. falciparum sporozoites marks the beginning of asymptomatic liver-stage infection. Within hepatocytes, parasites replicate massively as schizonts over 6–7 days before releasing thousands of merozoites that infect red blood cells. This stage is clinically silent but essential for establishing malaria infection. Glypican-3 (GPC3), a heparan sulfate proteoglycan on hepatocyte surfaces, has been identified as a putative receptor mediating P. falciparum sporozoite invasion. Certain subtypes of human hepatocytes expressing glutamine synthetase (hGS) are more permissive for parasite development. Understanding how P. falciparum invades and develops within human hepatocytes is crucial for: developing preerythrocytic vaccines aimed at blocking this stage, identifying novel drug targets among host receptors or metabolic pathways required by the parasite, improving in vitro models for antimalarial drug screening.
