Plasmodium falciparum translationally controlled tumor protein (PfTCTP) is a highly conserved, multifunctional eukaryotic protein homologous to human TCTP, secreted by the malaria parasite into host serum during infection. PfTCTP is involved in calcium ion binding and microtubule stabilization, playing roles in parasite growth, cell cycle regulation, and immune modulation. Structurally, PfTCTP shares significant similarities with human TCTP, but exhibits key differences in secondary structure and the G-protein binding region. Functionally, PfTCTP can be incorporated into host B cells and modulate their proliferation. It interacts directly with the antimalarial drug artemisinin, a feature that may be involved in the drug’s mechanism against the parasite. The release of PfTCTP during malaria infection enhances host basophil reactivity and contributes to histamine release and inflammatory responses. Elevated plasma levels of PfTCTP are observed in malaria patients, suggesting a role in disease pathogenesis and potential as a biomarker, but also posing challenges for immunomodulatory therapeutic approaches due to cross-reactivity and immune mimicry[1][2][3][4][5][7][8].
