Overview

Plasmodium parasite infectivity refers to the multi-stage biological process by which malaria-causing parasites establish an infection within a vertebrate host or a mosquito vector. This process encompasses several critical phases, including the initial inoculation of sporozoites by a mosquito, their migration to and invasion of liver cells, and the subsequent invasion of red blood cells by merozoites. Each of these steps relies on a complex interplay between parasite surface proteins—such as the Circumsporozoite Protein (CSP) and Apical Membrane Antigen 1 (AMA1)—and host cell receptors. In the context of drug and vaccine development, 'infectivity' is frequently targeted as a phenotypic outcome rather than a single molecular entity. Therapeutics aimed at reducing infectivity include transmission-blocking vaccines (TBVs) that prevent the parasite from maturing in the mosquito and pre-erythrocytic vaccines like RTS,S that target the parasite before it can cause clinical illness. Understanding the molecular determinants of infectivity is essential for developing interventions that can achieve malaria elimination by breaking the cycle of transmission.
