Plasmodium spp. DNA is the hereditary material of the Plasmodium parasites, the causative agents of malaria in humans. It serves as a critical therapeutic target for several antimalarial compounds that disrupt the parasite's ability to replicate and express essential genes. Some drugs, such as pentamidine, bind specifically to the minor groove of the DNA, while others like chloroquine and mepacrine can intercalate into the double helix, interfering with topoisomerase activity and DNA synthesis (PMID: 6101413, PMID: 15591011). Additionally, artemisinin derivatives have been shown to induce oxidative damage to the parasite's DNA, leading to cell death (PMID: 26690930). Beyond its role as a drug target, Plasmodium DNA is the primary biomarker used in molecular diagnostics, where PCR-based amplification of specific sequences allows for the sensitive detection and species-level identification of the parasite in clinical samples (WHO Malaria Terminology). The high AT-content of the P. falciparum genome presents both a challenge and an opportunity for the design of selective DNA-binding agents that distinguish between parasite and host genetic material.
