Overview

Plasmodium vivax ookinete surface protein 25 (Pvs25) is a cysteine-rich, surface-exposed protein expressed on the zygote and ookinete stages of P. vivax in the mosquito midgut. Structurally, Pvs25 contains four EGF-like domains stabilized by 11 disulfide bonds. It plays a key role in the parasite's development within mosquitoes and is absent from human blood stages. Pvs25 is a leading candidate for malaria transmission-blocking vaccines: antibodies raised against Pvs25 can inhibit parasite development in the mosquito, thus preventing onward transmission to humans. Pvs25 is highly conserved, but recombinant expression is technically challenging due to its complex disulfide-bonded structure. Previous protein subunit vaccines have shown potent transmission-blocking immunity in animal models, but immunogenicity in humans was suboptimal, driving interest in novel platforms, like mRNA vaccines, to improve efficacy[1][3][4][5][7][8].
