Overview

Plastin-1 (PLS1), also called fimbrin or I-plastin, is an intestine-specific actin-bundling and cross-linking protein predominantly expressed in the microvilli of intestinal and kidney epithelial cells and in hair cell stereocilia. It belongs to a family of cytoskeletal proteins critical for assembling and stabilizing tightly bundled actin filaments, which are essential for the structure and resilience of membrane protrusions such as microvilli and stereocilia. Loss of PLS1 affects intestinal brush border morphology, leading to increased epithelial cell fragility and susceptibility to inflammation, and mutations in the gene are associated with hereditary hearing loss. There are no known drugs directly targeting PLS1, and it is not considered a major therapeutic target at this time.
