Overview

Platelet calcium signaling is a fundamental biological process that orchestrates platelet activation, aggregation, and procoagulant activity in response to vascular injury [1, 2]. This complex signaling network is initiated by the activation of various surface receptors, including G protein-coupled receptors (GPCRs) like P2Y1, P2Y12, and PAR1/4, as well as immunoreceptor tyrosine-based activation motif (ITAM)-linked receptors such as GPVI [1, 4]. These receptors trigger the activation of phospholipase C (PLC) isoforms, leading to the production of inositol 1,4,5-trisphosphate (IP3), which releases calcium from the dense tubular system [2, 9]. The resulting depletion of intracellular stores activates store-operated calcium entry (SOCE) through the STIM1/Orai1 complex, causing a sustained rise in cytosolic calcium levels [2, 10]. This elevation is critical for downstream events such as integrin alphaIIbbeta3 activation, granule secretion, and the exposure of procoagulant phosphatidylserine [7, 9]. Dysregulation of this pathway is a major driver of arterial thrombosis, leading to conditions such as myocardial infarction and stroke [4, 5]. Consequently, components of the platelet calcium signaling pathway are primary targets for antiplatelet drugs, including P2Y12 inhibitors and PAR1 antagonists, which aim to prevent pathological thrombus formation while minimizing bleeding risks [5, 6].
