Platelet cyclooxygenase-1 (COX-1) is an enzyme predominantly expressed in platelets, where it plays a central role in the biosynthesis of thromboxane A2 (TXA2), a potent mediator of platelet aggregation and vasoconstriction. It catalyzes the conversion of arachidonic acid into prostaglandin H2 (PGH2), which is then converted into TXA2. Aspirin irreversibly inhibits COX-1, reducing platelet aggregation and providing antithrombotic effects. Inadequate inhibition or resistance to aspirin can occur, leading to persistent thromboxane generation. COX-1 also contributes to inflammation-related vascular diseases.
