Overview

Platelet-derived growth factor C (PDGF-C) is a 345-amino acid secreted protein encoded by the PDGFC gene, and is part of the PDGF/VEGF family of growth factors[1][7]. It functions predominantly as a mitogenic and chemotactic factor for cells of mesenchymal origin, playing essential roles in embryonic development (notably the heart, central nervous system, ear, kidney, and craniofacial structures such as the palate), tissue repair, fibrosis, and angiogenesis[2][5][7]. PDGF-C is synthesized as a latent, inactive precursor and requires proteolytic activation for biological activity, primarily binding to and activating the platelet-derived growth factor receptor alpha (PDGFR-α)[1]. Its dysregulation is implicated in pathological processes such as cancer, fibrosis, cardiovascular disease, and congenital anomalies including cleft lip and palate[1][2]. Because of its diverse roles in development and disease, PDGF-C and its signaling pathway represent attractive but challenging targets for therapeutic intervention, particularly in oncology and fibrotic disorders[2][5].
