Overview

Platelet-derived growth factor receptors (PDGFRs) are cell surface receptor tyrosine kinases that play a central role in regulating cell proliferation, differentiation, growth, and development. They are especially important in connective tissue cells such as fibroblasts and smooth muscle cells. There are two main types: PDGFRα and PDGFRβ. Ligand binding induces receptor dimerization and autophosphorylation, triggering downstream signaling cascades involved in cell proliferation, migration, survival, and differentiation. Aberrant activation or overexpression/mutation of PDGFRs has been implicated in various diseases including cancers, fibrotic disorders, and atherosclerosis, making them important therapeutic targets.
