Overview

The platelet glycoprotein Ib receptor complex, commonly referred to as the GPIb-IX-V complex, is a major membrane receptor found exclusively on the surface of platelets. It plays a central role in hemostasis by mediating the initial adhesion of platelets to sites of vascular injury, primarily through its interaction with von Willebrand factor (VWF) exposed on damaged subendothelium. Deficiency or dysfunction leads to Bernard–Soulier syndrome, a rare congenital bleeding disorder characterized by macrothrombocytopenia. Gain-of-function mutations can cause platelet-type von Willebrand disease. Autoantibodies against this complex are implicated in immune thrombocytopenic purpura. Involvement has been noted in thrombosis (e.g., stroke/myocardial infarction), metastasis facilitation for certain cancers, regulation of thrombopoiesis/platelet clearance, and possibly inflammatory responses.
