Platinum-DNA adduct formation refers to the covalent binding of platinum-based anticancer drugs (such as cisplatin, carboplatin, and oxaliplatin) to DNA. This process is central to the cytotoxic mechanism of these drugs, which are widely used in chemotherapy for various solid tumors. The platinum atom forms coordinate covalent bonds with nucleophilic sites on DNA bases—primarily the N7 position of guanine—resulting in a range of DNA lesions that disrupt cellular processes and trigger cell death.
