Overview

Pleckstrin and Sec7 domain containing 2 (PSD2) is a protein-coding gene that encodes a protein characterized by a pleckstrin homology (PH) domain and a Sec7 domain, with function as a guanine nucleotide exchange factor (GEF)[1][2][5][6]. PSD2 is involved in the regulation of ADP-ribosylation factor (ARF) protein signal transduction, phospholipid binding, and is implicated in processes such as cytoskeletal remodeling and membrane trafficking[2]. It is predicted to be located in intracellular structures such as the cleavage furrow and ruffle membrane, and is suggested to play a role at glutamatergic synapses and postsynaptic regions[2]. Diseases associated with mutations or dysregulation of PSD2 include spastic paraplegia and Gerstmann syndrome[2]. Its promoter can undergo DNA methylation and histone modifications, resulting in gene silencing. In some cancers, reduced PSD2 expression (potentially by epigenetic silencing) may contribute to tumor progression, suggesting a possible tumor suppressor role[4]. No direct drug interactions are currently established, but its modulation is being investigated for biomarker and therapeutic potential in oncology[4].
