Overview

Pleckstrin homology and FYVE domain-containing protein 1 (PLEKHF1, also known as Phafin-1) is a cytoplasmic protein involved in regulation of endosomal and lysosomal dynamics, particularly mediating autophagy and caspase-independent cell death. It contains both PH (pleckstrin homology) and FYVE domains, which confer lipid binding and membrane targeting functions. PLEKHF1 localizes to endosomes and lysosomes, where it can induce apoptosis via the lysosomal-mitochondrial pathway by promoting lysosomal membrane permeabilization and engaging downstream effectors for cell death. It also participates in the organization of the endosomal compartment and autophagosome formation, in part through Rab7 signaling. While it plays key roles in intracellular trafficking, autophagy, and cell death, there is currently no evidence that PLEKHF1 is a direct therapeutic target or that drugs directly interact with it[1][2][4][5].
