Pleckstrin homology and RhoGEF domain containing G3 (PLEKHG3) is a cytosolic protein that functions as a Rho guanine nucleotide exchange factor (RhoGEF), selectively activating the small GTPases RAC1 and CDC42 to regulate actin polymerization and is essential for cell polarity and directional motility[1][2][3]. It contains a tandem Dbl homology (DH)–pleckstrin homology (PH) domain, alongside an actin-binding domain that allows it to bind newly polymerized actin filaments at the leading edge of migrating cells[1]. PLEKHG3 acts as a key mediator in positive feedback loops coordinating polarized cell migration, and its loss impairs cellular motility and polarity. Disease associations include cancer, centronuclear myopathy, and autism spectrum disorder[2].
