Overview

Pleckstrin homology domain containing A6 (PLEKHA6) is a member of the pleckstrin homology domain-containing protein family and acts as a scaffold/adaptor molecule involved in phosphoinositide signaling, cytoskeletal remodeling, and cell adhesion. PLEKHA6 is overexpressed in lung adenocarcinoma and functions as an oncogenic driver by promoting β-catenin stabilization and Wnt signaling, supporting cancer cell proliferation, migration, and immune evasion. Single-cell analysis highlights PLEKHA6's specific expression in tumor-associated macrophages, particularly those with immunosuppressive phenotypes, which may foster a tumor-promoting microenvironment. Experimental targeting of PLEKHA6 impairs tumor cell growth and mobility in vitro, and its expression correlates with poor prognosis in cancer patients, suggesting utility as a biomarker and emerging precision oncology target.
