Pleckstrin homology-like domain family A member 1 (PHLDA1) is an evolutionarily conserved, proline-histidine rich protein encoded by the PHLDA1 gene in humans[1][2][6]. It contains a pleckstrin homology-like (PHL) domain, which enables it to bind phosphatidylinositol lipids and possibly participate in cell signaling, vesicular trafficking, transcriptional activation, and cytoskeletal rearrangement[2][3]. PHLDA1 was first identified as a factor in Fas-mediated apoptosis and activation-induced cell death in T lymphocytes[2]. The protein exhibits widespread tissue expression, with subcellular localization varying between cytoplasmic, perinuclear, nucleolar, and nuclear compartments[2][4]. Functionally, PHLDA1 modulates multiple biological processes, including apoptosis (cell death), cell proliferation, migration, and differentiation, in a context-dependent manner[2][3]. Its expression is responsive to external stimuli such as estrogens, growth factors, and cellular stress, and it can impact the balance between pro-apoptotic and anti-apoptotic signaling[2][3]. In cancers, PHLDA1 may exhibit both tumor suppressor and oncogenic roles, with downregulation linked to melanoma progression and overexpression associated with tumor proliferation and migration in colorectal cancer[2][3][4]. PHLDA1 interacts with integrins ITGA2 and ITGA6 and regulates cell adhesion and migration[3]. Recent data also support its utility as a marker of epithelial stem cells in human intestine, where it helps distinguish cellular hierarchies in normal and neoplastic tissues[3]. However, it is not a classical receptor, enzyme, transporter, or GPCR target, and no clinically approved drugs are known to directly target PHLDA1. No evidence currently supports its direct use as a drug target or therapeutic receptor. Instead, it is primarily of interest as a functional biomarker and a modulator of cell death and proliferation in health and disease, particularly in oncology and epithelial biology[2][3][4][6].
