It contains multiple protein interaction domains, including pleckstrin homology (PH), MyTH4, and FERM domains, which are commonly involved in cytoskeletal linkage and membrane association, suggesting an adaptor or scaffold protein function. PLEKHH3 has been reported to localize in the extracellular space, but its precise biological roles are largely uncharacterized. Limited evidence links PLEKHH3 to certain cancers, such as tracheal tumors, but it is not currently considered a therapeutic target or established biomarker[3][5].
