Overview

Plexin-A4 is a single-pass transmembrane receptor primarily involved in semaphorin signaling, where it acts as a coreceptor for neuropilin and class 3 semaphorins such as Sema3A, Sema6A, and Sema6B[1][2][3]. It is abundantly expressed in the nervous system and mediates processes including axonal guidance, synaptic targeting, neural differentiation, and neural regeneration. Plexin-A4 signaling leads to cytoskeletal reorganization, affecting cell migration, axon pruning, dendritic branching, and vascular remodeling[1][3][4][7]. Plexin-A4 is also implicated in immune regulation and has been found to play a role in the pathogenesis of Alzheimer’s disease, partly through its interaction with clusterin (CLU)[5], and may serve as a therapeutic target in cancer and neurodegenerative diseases.
