Plexin-D1 receptor (PLXND1) is a single-pass transmembrane receptor belonging to the plexin family and is the sole member of the vertebrate-specific PlexinD subfamily. It serves primarily as a high-affinity receptor for class III semaphorins, especially Semaphorin 3E, and mediates a variety of signal transduction events crucial for embryonic development, including guidance of axons, vascular patterning, regulation of immune cell migration, and skeletal morphogenesis. Its intracellular domain features GTPase-activating (RasGAP) motifs, allowing direct interaction with, and regulation of, small GTP-binding proteins. Plexin-D1 signaling is implicated in cancer progression, particularly due to its overexpression in tumor vasculature and malignant cells, making it a target and potential biomarker for diagnosis and therapy. It also interacts functionally with adaptor proteins such as GIPC and participates in protein complexes involved in endocytosis and cytoskeletal rearrangement[1][2][3][4].
