PML-RARA regulated adaptor molecule 1 (PRAM1) is a cytoplasmic adaptor protein implicated in signal transduction during immune cell (especially neutrophil and T cell) activation and differentiation. Its expression is regulated during normal myelopoiesis and is induced by retinoic acid, but suppressed by the oncogenic fusion PML-RARα in acute promyelocytic leukemia. Functionally, PRAM1 is similar to the FYN binding protein FYB/SLAP-130, participates in integrin signaling in neutrophils, and helps coordinate integrin-dependent cell responses such as adhesion and reactive oxygen species (ROS) production. Loss of PRAM1 impairs neutrophil degranulation and integrin-mediated signaling. It has also been implicated in the retinoic acid-induced differentiation program suppressed in leukemogenesis, and interacts with signaling proteins like HIP-55 and TRIM27[1][2][3].
