Pneumococcal surface protein A (PspA) is a major choline-binding protein found on the surface of nearly all strains of Streptococcus pneumoniae. It is highly immunogenic and plays a critical role in pneumococcal virulence, immune evasion, and pathogenesis. PspA consists of three main domains: an N-terminal alpha-helical domain (αHD), a proline-rich domain (PRD), and a C-terminal choline-binding domain (CBD). It inhibits complement activation, protects against antimicrobial peptides, and acts as an adhesin. Antibodies raised against PspA can protect against S. pneumoniae, suggesting its potential as a vaccine antigen, though sequence diversity poses challenges.
