Overview

Pneumolysin is a pore-forming toxin and a major virulence factor produced by the Gram-positive bacterium Streptococcus pneumoniae. It binds to cholesterol in host cell membranes, leading to oligomerization and formation of large pores. Pore formation disrupts cellular homeostasis and contributes to pneumococcal disease pathogenesis by facilitating colonization, promoting invasion, interfering with immune responses, and causing tissue injury. It activates innate immunity through TLR4 and the NLRP3 inflammasome and is an attractive target for vaccine development or therapeutic intervention.
