Overview

POC1 centriolar protein A (POC1A) is a centrosomal protein characterized by an N-terminal WD40 domain and a C-terminal coiled-coil domain[3][4][6]. It is essential for the assembly, stability, and function of centrioles and the formation of cilia. POC1A participates in the early steps of centriole duplication as well as in regulating centriole length, thus ensuring proper cell division and maintaining genomic stability. Mutations in POC1A lead to short stature, onychodysplasia, facial dysmorphism, and hypotrichosis (SOFT) syndrome[3][4][1]. In cancer biology, especially gastric cancer, POC1A acts as a tumor suppressor, regulating the cell cycle, DNA replication, and likely contributing to immune infiltration within the tumor microenvironment[1]. Its decreased expression is associated with tumor progression and poorer outcomes, making it a potential prognostic biomarker in oncology[1]. There are currently no known drugs directly targeting POC1A.
