POC1 centriolar protein homolog B (POC1B) is a core component of the centriole and centrosome, characterized by an N-terminal WD40 repeat domain and a C-terminal coiled-coil region connected by a flexible linker.[1] It plays an essential role in the stability and assembly of centrioles and is required for appropriate ciliogenesis and cell cycle progression.[2][3] POC1B and its paralogue POC1A have partially redundant but also unique functions: loss of both severely disrupts centriole maturation and spindle formation, while specific depletion of POC1B perturbs cell proliferation and may affect mitotic progression via mitosis-specific phosphorylation.[3][4] POC1B interacts with other inner centriolar proteins, including POC5, FAM161A, and MDM1, forming part of the inner centriole scaffold that maintains structural integrity.[1] Dysfunction or mutation of POC1B can lead to cellular phenotypes such as loss of centriole integrity, defects in cell division, and is associated with ciliopathies and inherited retinal disorders in humans, but POC1B itself is not an established pharmacological target.[3][2][1]
