Overview

POC5 centriolar protein (POC5) is a cytoskeletal protein localizing to the centriole and centrosome. It is essential for the assembly and elongation of the distal half of centrioles, acting also as a negative regulator to prevent over-elongation[2][3][4][6]. POC5 plays a central role in the formation of the inner scaffold, thereby stabilizing centriolar structure and facilitating proper compartmentalization of other centriolar proteins including Centrin, FAM161A, MDM1, and CCDC15[1]. Its function is critical for maintaining the integrity of centrioles and the organization of associated proteins, and loss of POC5 leads to disruption of centriolar architecture and associated protein localization. Genetic mutations in POC5 are linked to human diseases, notably syndromic rod-cone dystrophy and other retinal diseases[2]. POC5 is not currently recognized as a therapeutic target, nor are there known interacting drugs, mechanisms of drug action, biomarkers, or safety concerns linked to pharmacological targeting. POC5 is a structural and organizational component protein, not a classical drug target such as a receptor, enzyme, transporter, or ion channel. It fits into the “Other” molecular classification for cytoskeletal and centriolar proteins.
