Overview

Podocalyxin (PODXL) is a single-pass transmembrane sialomucin and major component of the podocyte glycocalyx in the kidney glomerulus, where it maintains open filtration pathways between foot processes through negative charge repulsion. It belongs to the CD34 family, features extensive O- and N-linked glycosylation, and links to the actin cytoskeleton via ERM proteins. Beyond kidney function, it regulates apical domain formation, microvilli induction, cell migration, and invasion; elevated expression correlates with poor cancer outcomes, acting as a decoy receptor for galectin-3 to promote integrin-mediated motility. The protein is 605 amino acids long with a mass of approximately 65 kDa (higher due to glycosylation), including extracellular (33-500), transmembrane (501-521), and cytoplasmic (522-605) domains.
