Overview

POLH antisense RNA 1 (POLH-AS1) is a long non-coding RNA transcribed from the antisense strand of the POLH gene. It does not encode a protein but plays regulatory roles at the RNA level. POLH-AS1 is highly expressed in hepatocellular carcinoma (HCC), where its elevated expression associates with more advanced disease and poorer prognosis. Functionally, POLH-AS1 promotes HCC cell proliferation, migration, and invasion, and its inhibition reduces these malignant behaviors. POLH-AS1 also modulates several cell death pathways (such as necroptosis, ferroptosis, and cuproptosis) and is implicated in the regulation of the tumor immune microenvironment, correlating with immune infiltration and immune checkpoint gene expression. Its precise molecular mechanisms are still being investigated, but current evidence suggests POLH-AS1 acts as a key oncogenic lncRNA and a potential therapeutic and prognostic target in HCC.
