The Poliovirus Receptor (PVR), also known as CD155, is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. It functions in cell adhesion, immune regulation, viral entry (specifically for poliovirus), and tumor biology. It interacts with immune receptors such as CD226 (DNAM-1) and TIGIT, modulating NK and T cell activity. PVR expression is upregulated in various cancers, making it a potential biomarker and therapeutic target. Engineered polioviruses are being investigated as cancer therapies due to their tropism for high-PVR-expressing cancer cells.
