Overview

CD112, also known as poliovirus receptor-related 2 protein or nectin-2, is a cell surface adhesion molecule belonging to the immunoglobulin superfamily and functions as a type I transmembrane glycoprotein. It is broadly expressed in epithelial, endothelial, neuronal, and hematopoietic cells. CD112 plays major roles in cell–cell adhesion (especially at adherent junctions), immune regulation as a ligand for immune receptors like TIGIT, CD112R (PVRIG), and DNAM-1 (CD226), and as a mediator of viral entry for certain herpesviruses. High expression or altered regulation of CD112 has been linked to tumor progression, metastasis, and poor prognosis in various cancers. CD112 is considered a novel target for immune checkpoint inhibition strategies in cancer immunotherapy.
