Poliovirus receptor-related immunoglobulin domain-containing protein (CD112R) (CD112R)

Poliovirus receptor-related immunoglobulin domain-containing protein (PVRIG), commonly referred to as CD112R, is an inhibitory immune checkpoint receptor predominantly expressed on T cells and Natural Killer (NK) cells (UniProt: Q6DKI7). It belongs to the poliovirus receptor (PVR)-like family and specifically interacts with its ligand CD112 (PVRL2), which is frequently overexpressed on various tumor cells (PubMed: 27647348). Upon binding to CD112, CD112R transduces inhibitory signals that suppress the activation, proliferation, and cytokine production of cytotoxic lymphocytes, thereby contributing to tumor immune escape (PubMed: 31043417). Because the PVRIG pathway is distinct from and complementary to the TIGIT and PD-1 pathways, it has emerged as a promising therapeutic target in oncology (PubMed: 32103181). Therapeutic agents such as COM701 are monoclonal antibodies designed to block this interaction, aiming to restore the anti-tumor activity of the immune system (ClinicalTrials.gov: NCT03667716). Current clinical research focuses on using CD112R antagonists both as monotherapy and in combination with other checkpoint inhibitors to treat advanced solid tumors (PubMed: 34161144).
