The poliovirus replication complex is a specialized, virus-induced structure within infected host cells that orchestrates the replication of the poliovirus RNA genome. It is a dynamic assembly of viral proteins, newly synthesized viral RNA, and extensively remodeled host cell membranes. The core components include viral nonstructural proteins (notably 3A, 3AB, 3B/VPg), newly synthesized viral RNA, and host factors involved in membrane trafficking. Its primary function is to provide an optimal microenvironment for efficient synthesis of both negative-strand (template) and positive-strand (genomic) RNAs. Formation of these complexes leads to severe reorganization within the cell, ultimately causing cytopathology and cell death.
