Overview

The Poliovirus type 1, 2, 3 antigens are the serotype-specific capsid proteins present on the surface of the poliovirus particle, a non-enveloped virus containing a single-stranded positive-sense RNA genome. These antigens (PV-1, PV-2, PV-3) determine the serotype and antigenic distinctiveness of the viruses. They are critical targets for neutralizing antibodies and vaccines, with immune recognition largely mediated by conformational epitopes within the capsid. Interaction of these antigens with the cellular receptor CD155 is required for host cell entry, and antigenic variation explains differences in vaccine effectiveness and disease transmission. The antigens are used in both live attenuated and inactivated polio vaccines, and their presence in the blood as antibody targets serves as a biomarker for immunity. Therapeutic monoclonal antibodies can bind these antigens, though effectiveness may vary depending on the serotype and affinity. Vaccine-derived strains can occasionally lead to safety concerns in under-immunized populations, but polio vaccination remains the primary tool for global disease prevention.

Vaccine: Induction of protective neutralizing antibodies against the viral antigens to prevent infection. Monoclonal antibodies: Bind to antigenic sites on the viral capsid, overlapping the receptor-binding site for CD155, prevent viral entry and neutralize the virus
