The Poliovirus type 1 capsid protein VP1-VP4 complex, often abbreviated as VP1-VP4, is a key viral structural protein, capsid protein, and protein complex. Its primary biological functions include virus particle formation and stability, host cell receptor binding and viral entry, protection of the viral RNA genome, and serving as a critical antigen that elicits neutralizing antibodies. This complex is central to the pathogenesis of poliomyelitis, the disease caused by poliovirus. While this complex is the target of existing vaccines that induce immunity, there are no FDA-approved small molecule drugs specifically interacting with it, meaning there is no direct drug mechanism of action. Although neutralizing antibody titers against these proteins are used to assess vaccine efficacy, the complex itself is not utilized as a biomarker in clinical practice. Notable safety concerns associated with this complex, particularly in the context of vaccine development, include the potential for antigenic instability of empty capsids and the risk of reversion to neurovirulence when using live attenuated vaccines containing these components. Other names for this complex include Poliovirus type 1 major capsid proteins, Poliovirus structural proteins, Poliovirus virion coat proteins, and PV-1 VP0 (which is a precursor). It is also sometimes referred to as a "procapsid" or "pentameric assembly" during the process of virus assembly.
