Overview

Poliovirus type 3 inactivated virion antigens are the structural components of the type 3 poliovirus (typically the Saukett strain) that have been rendered non-infectious through chemical treatment, usually with formaldehyde (CDC, 2021). These antigens are a critical component of the Inactivated Poliovirus Vaccine (IPV), which is used globally to prevent poliomyelitis (WHO, 2022). The primary biological role of these antigens is to serve as a template for the host's immune system to recognize and develop a memory response against the wild-type virus (StatPearls, 2023). Upon administration, the antigens trigger the production of neutralizing antibodies that circulate in the blood, providing systemic immunity (FDA, 2020). This prevents the virus from spreading to the central nervous system and causing paralysis if the individual is later exposed to the live virus (PubMed, 2019). Unlike the oral poliovirus vaccine, the inactivated antigens cannot replicate or revert to a virulent form, making them essential for the final stages of global polio eradication (NIH, 2021).

Mechanism of action

The inactivated virion antigens act as exogenous antigens that are processed by antigen-presenting cells and presented to B and T lymphocytes, stimulating the production of serotype-specific neutralizing antibodies (IgG) that prevent the virus from entering the central nervous system (CDC, 2021; StatPearls, 2023).
