Overview

The Pollen allergen-specific immune receptor complex represents the molecular interface of the adaptive immune response to pollen allergens, involving the interaction between pollen-derived peptides presented on Major Histocompatibility Complex class II (MHC-II) and their cognate T-cell receptors (TCRs), alongside the recognition of intact allergens by B-cell receptors (BCRs). In sensitized individuals, this axis drives a Th2-polarized immune response, leading to the production of allergen-specific IgE and subsequent mast cell degranulation (Larche et al., 2006). Therapeutic strategies targeting this complex aim to induce immunological tolerance or "desensitization" through Allergen-Specific Immunotherapy (AIT), which modifies T-cell behavior from a pro-inflammatory Th2 state to a regulatory (Treg) or Th1 state (Akdis & Akdis, 2014). Modern approaches also include peptide-based vaccines designed to bind MHC-II and engage TCRs without cross-linking IgE on mast cells, thereby reducing the risk of anaphylaxis (Shamji & Durham, 2017). Understanding this complex is crucial for developing precision medicines for allergic rhinitis and asthma, as it sits at the apex of the allergic cascade.
