Poly(adenosine-diphosphate-ribose) polymerase 1 (PARP1) is a multifunctional nuclear enzyme that plays a crucial role in maintaining genome integrity, responding to cellular stress, and regulating transcription. It is a key player in DNA damage repair, particularly in response to single-strand and double-strand breaks. PARP1's activity is essential for base excision repair, nucleotide excision repair, non-homologous end joining, and homologous recombination repair. Its inhibition is therapeutically exploited in cancers with defective homologous recombination pathways.
